Product Description

S-Adenosyl methionine (S-Adenosyl methionine, SAM) is an auxiliary substrate involved in the methyl transfer reaction, which in the body is composed of adenosine triphosphate and methionine in the methionine adenosyltransferase under the action of synthesis. SAM is prepared in a solution of 10 mM HCl, 10% Ethanol, pH4 at 25℃.

This product is produced in accordance with GMP process requirements and provided in liquid form.

图1用40μg/ml的AF488抗SAH 301-3(Cat#MAF00301)和8μg/ml的AF647抗SAM 118-6(Cat#MAF00201)对HepG2和L02细胞进行免疫荧光(IF)染色,然后进行DAPI染色,并在激光扫描共聚焦显微镜(蔡司LSM 780)下拍摄。(A1-A4)正常肝细胞系L02细胞在含有10%FBS的RPMI 1640中培养40h。
不同观点如下:DAPI(A1);AF488用于SAH(A2);SAM的AF647(A3);所有三个荧光信号的重叠(A4)。(B1-B4)肝细胞癌细胞系HepG2细胞培养40小时。不同观点如下:DAPI(B1);AF488用于SAH(B2);用于SAM(B3)的AF647;所有三个荧光信号的重叠(B4)。在L02和HepG2细胞(x630)之间,SAM和SAH的表达模式不同。

 

图2:36μg/ml下Alexa Fluor9488结合抗SAH抗体301-3(Cat#MAF00301)和4.5 ug/ml下Alexa Fluorm 647结合抗SAM抗体118-6(Cat#MAF00201)双重染色的L02(A1-A3)和HepG2(B1-B3)细胞的流式细胞术。
颜色图例:
橙色:空白;蓝色:使用核固定/通透性缓冲液(eBioscience 00-5523 FoxP3uTF染色缓冲液组);红色:使用细胞内固定/渗透缓冲液(eBioscience 00-8824)。用细胞内固定/通透性缓冲液(A2,B2)或核固定/通透性缓冲液(A3,B3)固定并通透性融合细胞(在含10%FBS的RPMI 1640中培养48小时),然后用上述抗体双重染色。使用BD-FACSCalibur流式细胞仪分析细胞。
SAM在L02细胞中的表达高于HepG2细胞。SAM和SAH都广泛表达,但相当动态。两种细胞中SAM的水平均高于SAH。
