因子X是一种维生素K依赖性蛋白酶原，在肝脏中合成，并在血浆中以通过二硫键连接的两链分子（1,2）的形式循环。在分泌到血浆中之前，翻译后修饰产生位于NH2末端轻链内的11个γ-羧基谷氨酸（gla）残基和单个b-羟基天冬氨酸残基。轻链还包含两个表皮生长因子（EGF）同源域。因子X的COOH末端重链包含大部分碳水化合物部分以及潜在的丝氨酸蛋白酶结构域。因子X的激活被内在因子Xase复合物（因子IXa，因子VIIIa，细胞表面和钙离子）或外在因子Xase复合物（因子VIIa，组织因子，细胞表面和钙离子）催化。通过任何一种复合物激活人因子X均会导致COOH末端重链的Arg52-Ile53裂解，并随后释放52个氨基酸的激活糖肽。然后，因子Xa充当凝血酶原酶复合物的酶成分，该酶原负责凝血酶原向凝血酶的快速转化。gla残基使因子X / Xa以钙依赖性方式结合磷脂（即细胞表面）。凝血酶原酶复合物组装的要求。一个EGF同源结构域包含一个Ca2 +结合位点，该位点充当将EGF和GLA结构域彼此折叠的铰链（12）。分子的该区域参与细胞结合结构域的识别。

通过常规技术（3）和免疫亲和色谱（4）的结合，从新鲜的冷冻人血浆中分离出人因子X。除了标准的人因子X制剂外，还提供无Gla域的人因子X。使用Bajaj等人报道的方法的改进方法从新鲜牛血浆中分离牛X因子。（5,6）。纯化的酶原以50％（体积/体积）甘油/ H2O的形式提供，应储存在-20oC下。通过SDS-PAGE分析确定纯度，并在X因子凝血测定中测量活性。

激活过程中被因子Xase蛋白水解切割的位点。

基于ANSN的植物底物
的一般结构举例说明了基于6-氨基-1-萘磺酰胺基（ANSN）的荧光底物的一般结构。在本化合物家族中，R 1是三肽，其COOH-末端残基通常是精氨酸。R 2和R 3可以是氢，烷基，芳基或环烷基。虚线表示各种丝氨酸蛋白酶的水解位点。

包含荧光报告基团6-氨基-1-萘-磺酰胺（ANSN）的底物是用于监测各种丝氨酸蛋白酶（1-12）的酶活性的有用化合物。在这类化合物中，ANSN报告基团（在R1位置）连接至短三肽序列。设计肽序列以优化酶和底物之间的相互作用。可以添加到R2和R3位置的其他组分反映了P'亚位位置的变化，并且通常影响底物的动力学参数。有效底物的化合物在三肽和ANSN基团之间水解。一旦从肽部分上切下，ANSN基团的相对荧光显示出约1000倍的增加。

下一页确定的动力学性质将有助于选择合适的底物。事实证明，ANSN底物对因子VIIa（1-5,7,9,10,12）的分析特别有用。尽管仅因子VIIa的底物水解速率相对较慢，但将组织因子掺入测定系统后，几种化合物（例如化合物SN-17a）的kcat表现出较大变化。

使用信息：
基于ANSN的底物在DMSO中以10mM储备液的形式提供，通常可用于分析浓度范围为400 nM至低至50pM的酶。ANSN底物通常以50至100 µM的最终浓度使用，并可以在大多数生理缓冲液（即TBS，HBS，PBS等）中稀释至该最终浓度。反应（荧光的相对变化）可以使用标准荧光计或荧光板读数器进行监测。可以将仪器设置在以下波长下使用动力学或终点模式（激发= 352nm，发射= 470nm）。通过在发射光束中使用390至450 nm长通截止滤光片，可以大程度地减少光伪影。

DMSO中的底物储备溶液应在4°C或更低的温度下冷冻保存，并应避光。在这些条件下，这些化合物将保持稳定超过一年。

的结构域表示蛋白质S的结构域结构，其中：GLA =含有γ-羧基谷氨酸残基的区域，EGF =含有与人表皮生长因子同源的序列的区域，TSR =凝血酶敏感区域，？=未知功能区域，取代维生素K依赖性丝氨酸蛋白酶中发现的催化三联体。

蛋白质S是单链维生素K依赖性蛋白质，被认为在凝血和补体级联中起作用（1,2）。在血浆中循环的蛋白质S的大约60％与C4b结合蛋白（C4BP）复合。已经表明，g-羧基谷氨酸（gla）依赖性蛋白S与带负电荷的磷脂的结合可以起作用以在损伤后将C4BP浓缩在细胞表面。

在凝血系统中，蛋白质S起抗凝血辅因子蛋白的作用。在Ca2 +和磷脂囊泡（Kd = 6×10-9M）（3）存在下，活化蛋白C（APC）与蛋白S形成1：1的化学计量复合物。在蛋白质S的存在下，在血浆和未刺激的血小板表面观察到因子Va的速率和APC的因子VIIIa失活的适度增加（3-10倍）。与C4BP结合的蛋白S不具有APC辅因子活性。近，已经描述了增强蛋白S活性的另外的结合蛋白（4）。已经提出凝血酶对蛋白S的蛋白水解失活作为该系统中的调节机制。通过凝血酶的单次切割通过去除含有gla结构域的NH 2末端肽（Mr = 8000）来消除蛋白S辅因子活性。

蛋白质S的结构域结构类似于其他维生素K依赖性凝血因子的结构域结构，除了蛋白质S不具有催化三联体。蛋白S是单链蛋白，在NH2末端结构域中含有10个gla残基，在4个表皮生长因子（EGF）结构域中含有10个gla残基。

如Jenny等所述，通过常规方法（9）和免疫亲和层析的组合从新鲜冷冻血浆中分离人蛋白S. （5）。纯化的蛋白质S以50％（体积/体积）甘油/ H 2 O供应，应储存在-20℃。通过SDS-PAGE分析确定纯度。
