产品描述

   成纤维生长因子7(FGF-7),也称为角质形成细胞生长因子 (KGF) ,是成纤维细胞生长因子 (FGF) 家族的成员。在胚胎发育、细胞增殖和细胞分化的调控中发挥重要作用。正常分枝形态发生所必需的,对角质形成细胞有活性的生长因子。可能是正常上皮细胞增殖的主要旁分泌效应物。在大肠杆菌中过表达的重组KGF活性约为天然蛋白的10倍,可能与重组KGF未被糖基化有关。KGF,122-132AA段是KGF受体的特异性结合位点。KGF由各种来源的间质细胞分泌,与肝素有较强的亲和力,其受体KGFR属于蛋白酪氨酸激酶受体家族,其主要分布于上皮细胞,KGF与靶细胞膜上的受体KGFR特异性结合后,促使受体自身磷酸化,从而启动细胞内信号级联反应,进而发挥多种生物学功能:参与组织器官发育、促进细胞增殖及组织损伤修复等。

样品采集和存储

请注意您感兴趣的套件推荐哪些样品类型。

血清

使用血清分离管,让样品在室温下凝固2小时或在4℃下过夜,然后以约1000×g离心20分钟。立即测定新鲜制备的血清,或将样品分装在-20℃或-80℃下备用。避免反复冻/融循环。

等离子体

使用EDTA或肝素作为抗凝血剂收集血浆。在收集后30分钟内,在2-8℃下以1000×g离心样品15分钟。立即去除血浆并进行检测,或将样品保存在-20℃或-80℃的等分试样中以备后用。避免反复冻/融循环。

组织匀浆

组织匀浆的制备将根据组织类型而变化。通常,组织应在冰冷的PBS(0.02mol / L,pH 7.0-7.2)中*冲洗以除去过量的血液并在均质化前称重。将组织切成小块并在冰上用玻璃匀浆器在5-10mL PBS中均化(Micro Tissue Grinders也起作用)。应使用超声波细胞破碎器对所得悬浮液进行超声处理,或进行两次冻融循环以进一步破坏细胞膜。之后,匀浆应以5000×g离心5分钟。立即取出上清液并进行分析,或等分试样并在≤-20℃下储存。

细胞裂解物

根据以下说明,在测定前必须裂解细胞:

  • 应用胰蛋白酶分离贴壁细胞,然后通过离心收集(悬浮细胞可通过直接离心收集)。
  • 在冷PBS中洗涤细胞三次。

注意:                                                

  • 样品在5天内可以被存储在4使用Ô C,否则样品必须储存在-20 Ô C(≤1月)或-80 Ô C(≤2个月),以避免生物活性和/或污染的损失。
  • 样品溶血会影响结果; 不会检测到用作样本的溶血标本。
  • 进行测定时,将样品置于室温下。

试剂准备

*使用前将所有试剂盒组分和样品置于室温(18-25 o C)。

标准 

用标准稀释剂重新配制标准品,在室温下保持10分钟,轻轻摇动(不要起泡)。有关储备溶液中标准浓度的信息,请参阅手册。准备7管含有0.5mL标准稀释剂的试管,并使用稀释的标准品生产双稀释系列。在下次转移之前*混合每个管。

检测试剂A和检测试剂B.

传统的ELISA试剂盒:使用前短暂旋转或离心检测A和检测B溶液。分别用Assay Diluent A和B稀释至工作浓度(1:100)。

即用型ELISA试剂盒:检测溶液A和B的浓度已经正确,无需进一步稀释。

洗液 

用580mL去离子水或蒸馏水稀释20mL洗涤液浓缩物(30×),制备600mL洗涤液(1×)。

TMB底物 

用灭菌的吸头吸取所需剂量的溶液。不要将残留的溶液倒回到小瓶中。

注意:

  • 不要直接在孔中进行连续稀释。
  • 在进行测定后15分钟内制备标准品。不要直接在37 o C 溶解试剂。
  • 检测试剂A和B是粘性溶液,因此慢慢移液它们以减少体积误差。
  • 传统的ELISA试剂盒:根据说明小心地重新构建标准品或使用检测试剂A和B,避免起泡并轻轻混合直至晶体*溶解。为了大限度地减少移液造成的不性,请使用小体积并确保移液器已校准。建议一次移液10μL以上以确保准确性。                 
  • 即用型ELISA试剂盒:重组标准品只能使用一次
  • 重构的标准品,检测试剂A和检测试剂B只能使用一次
  • 如果在洗涤液浓缩物(30×)中形成晶体,则温热至室温并轻轻混合直至晶体*溶解。
  • 试剂制备过程中使用的任何污染水或容器都会影响检测结果。

样品制备

  • Reddot Biotech仅对试剂盒本身负责,而不是对试验过程中消耗的样品负责。实验者应该在开始实验之前总是计算所需的样品总量。
  • 实验者还应该在开始之前预测大致的浓度。如果预期值不在标准曲线范围内,建议确定样品的宜稀释度。
  • 如果您使用的样品类型未在手册中指出,则应在开始实际实验之前进行初步实验以确保试剂盒的有效性。
  • 建议不要使用化学裂解缓冲液来制备组织或细胞提取样品,因为它可能会由于裂解缓冲液的含量而导致意外的反应和结果。
  • 由于其他来源的抗原与我们试剂盒中使用的抗体之间可能存在错配,因此不建议使用其他制造商生产的天然或重组蛋白,因为我们的试剂盒可能无法识别它们。

分析程序

用于夹心ELISA试剂盒

注意:

  • 孵育:为了确保准确的结果,需要在孵育步骤中适当粘附板密封剂。在孵化步骤之间不允许孔长时间未被覆盖。一旦将试剂添加到孔条中,在测定期间不要让条带干燥。必须控制孵育时间和温度。
  • 洗涤:洗涤程序至关重要。在每个步骤中*去除液体对于试剂盒的良好性能是*的。后一次清洗后,通过抽吸或倾析除去任何残留的清洗液,并去除板底部的任何水滴或指纹。洗涤不足将导致精度差和吸光度读数错误升高。
  • 控制反应时间:观察加入TMB底物后的颜色变化(例如每10分钟观察一次),如果颜色太深,则提前加入终止溶液以避免过强的反应,这将导致吸光度读数不准确。
  • TMB底物  很容易被污染。保护它免受光线和物理污染。 
  • 环境湿度可能对从试剂盒获得的结果产生影响。如果您工厂的湿度低于60%,建议使用加湿器。

 

测试原理

用于夹心ELISA试剂盒

该试剂盒中提供的微量滴定板预先涂有对靶抗原特异的抗体。然后将标准品或样品加入适当的微量滴定板孔中,用对靶抗原特异的生物素缀合的抗体制剂。接下来,将与辣根过氧化物酶(HRP)缀合的抗生物素蛋白加入每个微孔板孔中并孵育。在添加TMB底物溶液后,仅含有靶抗原,生物素缀合的抗体和酶缀合的抗生物素蛋白的孔将显示颜色变化。通过添加硫酸溶液终止酶 – 底物反应,并且在450nm±10nm的波长下通过分光光度法测量颜色变化。然后通过比较样品的OD与标准曲线确定样品中靶抗原的浓度。

适用于竞争性ELISA试剂盒

该测定采用竞争性抑制酶免疫测定技术。将靶抗原特异性的单克隆抗体预先包被在微孔板上。在生物素标记的靶抗原和未标记的靶抗原(标准品或样品)之间启动竞争性抑制反应,其中预涂的抗体对靶抗原具有特异性。温育后,洗去未结合的缀合物。接下来,将与辣根过氧化物酶(HRP)缀合的抗生物素蛋白加入每个微孔板孔中并孵育。结合的HRP缀合物的量是反向正比于样品中的靶抗原的浓度。加入底物溶液后,显色的强度与样品中靶抗原的浓度成比例。

灵敏度

该测定的灵敏度或检测下限(LLD)被定义为可以与零区分的低蛋白质浓度。通过将两个标准偏差加到二十个零标准重复的平均光密度值并计算相应的浓度来确定。

夹心ELISA试剂盒的典型数据

为了使计算更容易,我们绘制标准的OD值(X轴)与标准的已知浓度(Y轴),尽管浓度是自变量,OD值是因变量。然而,标准曲线的OD值可以根据测定性能的条件(例如操作者,移液技术,洗涤技术或温度效应)而变化,建议绘制数据的对数以建立每个测试的标准曲线。有关典型的标准曲线,请参阅套件中包含的手册。

确定所有套件精度的一般方法如下所述。可根据要求提供个人批量套件的规格。

  • 测定内精密度(测定内的度):分别在一个平板上测试3个具有低,中和高水平指数的样品20次。
  • 批间精密度(测定之间的度):在3个不同的平板上测试3个具有低,中和高水平指数的样品,每个平板8个重复。

稳定性

我们的ELISA试剂盒的稳定性取决于活性丧失的速率。在适当的储存条件下,我们的试剂盒的失效率在失效日期内通常低于5%。

为尽量减少外部对我们套件性能的影响,应始终控制操作程序和实验室条件。需要特别注意的因素包括:室温,空气湿度和培养箱温度。还强烈建议整个测定由相同的实验者从开始到结束进行。

Biological Activity Fully biologically active when compared to standard. The ED50 as determined by thymidine uptake assay using FGF-receptors transfected BaF3 cells is less than 0.5ng/ml, corresponding to a specific activity of >2.0×106IU/mg.
Physical Appearance Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation Lyophilized from a 0.2μm filtered concentrated solution in 2X PBS, 600mM NaCl, pH7.4, 1mM mercaptoethanol.
Endotoxin Less than 1EU/μg of rMuKGF-2/FGF-10 as determined by LAL method.
Reconstitution We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤-20℃. Further dilutions should be made in appropriate buffered solutions.

Background Fibroblast growth factor 10 belongs to the fibroblast growth factor (FGF) family, which is involved in a variety of biological processes such as embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. Like most other FGF family members, FGF-10 also has a heparin-binding domain and plays an important role in the regulation of embryonic development, cell proliferation and cell differentiation. In addition, FGF-10 may take parts in wound healing and is required for normal branching morphogenesis. Recombinant murine FGF-10 contains a 173 amino acids and it shares 93% and 90%a.a. sequence identity with human and rat FGF-10.

保存条件:
-20℃或更低温度保存,至少一年有效。由于蛋白的每次冻融均会引起部分失活,所以首次配制成相应浓度的储存液后(请根据产品简介中Reconstitution一栏的信息配制储存液),须分装后-20℃或更低温度冻存,以避免反复冻融。
注意事项:
由于有些塑料管壁对某些蛋白有较强的吸附作用,溶液中的蛋白很容易粘附在管壁上,并且粘附后的蛋白很难与管壁分离。而载体蛋白(Carrier protein,如0.1% BSA等)的主要作用是预先封闭塑料管壁上的蛋白结合位点,使细胞因子或重组蛋白不会粘附于管壁。所以一定要使用产品简介中Reconstitution一栏的信息配制储存液。

使用说明:
1. 收到产品后请立即按照说明书推荐的条件保存。除非特别注明,碧云天相关产品均为冻干粉,由于微量的蛋白在冻干过程中沉积在管内,形成很薄或不可见的蛋白层,所以在打开管盖前,我们建议在离心机中约8,000-12,000g离心10-30秒,使附着在管盖或管壁上的蛋白聚集于管底。
2. 请根据实验目的并按照产品简介中Reconstitution一栏中的信息配制储存液。大多数细胞因子或重组蛋白的冻干粉是非常容易溶解的,一般用移液枪的枪头轻吹几下或者轻轻摇晃瓶子,即可使细胞因子或重组蛋白完全溶解。请勿用vortex剧烈振荡,以免蛋白变性而失活。
3. 具体的最佳工作浓度请自行参考相关文献,或者根据实验目的,以及特定细胞和动物,通过实验进行摸索和优化。

二、试剂盒组成

1) FGF-23抗体包被微孔板(FGF-23板)

用抗FGF-23的鼠单克隆抗体包被微孔板96孔微孔板。  

  2) HRP标记的FGF-23抗体(FGF-23缀合物)

12mL与辣根过氧化物酶缀合的FGF-23的鼠单克隆抗体与防腐剂混合。该试剂应储存在2-10℃并避光。

  3) FGF-23校准液(STD1~std7)

用防腐剂缓冲FGF-23的FGF-23标准品(Std1-Std7)12mL(Std1)和0.5mL(Std2-Std7)。7瓶含有防腐剂的缓冲碱的FGF-23。  

  4) 测定稀释剂(Assay Diluent)

12 mL缓冲液。  

  5) 显色剂(底物)

12mL四甲基联苯胺TMBZ)与过氧化氢脲。  

  6) FGF-23清洗缓冲液(10倍浓缩)

53毫升缓冲表面活性剂与防腐剂的10倍浓缩溶液。使用前,用去离子水将内容物稀释至10倍并充分混合。  

  7) 终止液(终止液)

12 mL 0.5 mol / L硫酸。  

  8) 封板器

试剂盒中包含密封板三。  

       

    * 这些试剂应在黑暗中储存在2-10℃。使用前将所有试剂置于室温下。  

     

三、试剂盒检测原理

FGF-23 ELISA试剂盒是用于测量血清中FGF-23的双位点酶联免疫吸附测定试剂盒。两种特异性鼠单克隆抗体结合全长FGF-23。其中一种抗体固定在微量滴定板上进行捕获。另一种抗体与HRP(辣根过氧化物酶)缀合用于检测。

在*个反应中,将含有FGF-23的样品与固定微量滴定孔的抗体一起培养。用抗体捕获样品中的FGF-23。在该反应结束时,洗涤该孔以去除未结合的FGF-23和其他组分。

在第二个反应中,将这种固定的FGF-23与HRP标记的抗体一起温育以形成“三明治”复合物:抗FGF-23抗体 – N端FGF-23C端-HRP标记的抗FGF-23抗体在该反应结束时,洗涤该孔以除去未结合的组分。

在酶反应中,将固定在孔上的夹心复合物与底物溶液一起孵育,然后用分光光度微量滴定板读数器测量。与孔结合的复合物的酶活性与样品中FGF-23的量成正比。通过绘制吸光度与FGF-23标准品各浓度的关系绘制标准曲线。从该曲线确定样品中FGF-23的浓度。

 

 

四、产品使用注意事项

1)

测试样品的储存条件是什么?

 

使用鲜的血清样本。如果您无法立即测试您的样品,请将其存放在-70°C以下。避免解冻/重新冷却循环。

在我们的实验室测试中,在冰箱中13天后,给定样品的活性降至86.6%。样品在-80°C下可保持稳定约三年,但在-20°C时稳定性不能得到保证。

我们已确认zui多六次解冻/再次冻结循环可能不会影响活动,但我们建议小批量冻结样本,以尽量缩短解冻/再次冻结周期。

2)

试剂盒检测的样本

 

虽然血浆和血清都可以使用FGF-23 ELISA试剂盒进行检测,但建议使用血清。

3)

试剂盒是否可以测试动物样本

 

FGF-23ELISA试剂盒可用于测量小鼠,大鼠或猴血FGF-23水平。

4)

试剂盒是否含有任何汞基防腐剂

 

FGF-23 ELISA试剂盒不使用水银防腐剂,如硫柳汞。标准溶液,稀释溶液,酶标抗体溶液和浓缩洗涤溶液使用Kathon TM CG作为防腐剂。

5)

操作预防措施

 

校准曲线必须为每次测试准备。

如果测试多个样品,则反应时间必须保持一致。

搅拌板时必须进行一级反应。搅拌前应密封盘子。

洗涤周期中必须遵守以下预防措施:

   不要将洗版溶液留在其中。

   一旦从井中充分移除洗涤液,立即进入下一步。

 

五、试剂使用方法

1)

在支架上准备足够数量的抗体涂层带。

2)

向孔中加入50μL检测稀释剂。

3)

向每个孔中加入50μLFGF-23标准品或样品,并用平板密封器盖住平板。

4)

在板式混合器上在室温下孵育平板2小时。

5)

拆卸封板器。吸取每孔中的样品。用300μL工作洗涤缓冲液洗涤每个孔4次,并*吸出内容物。

6)

向每个孔中加入100μL的FGF-23结合物并用封板器覆盖该板。

7)

在平板混合器上在室温下孵育平板1小时。

8)

拆卸封板器。吸取每孔中的样品。每孔用300μL工作洗涤缓冲液洗涤4次,并将其*吸出。

9)

向每个孔中加入100μL底物,并用平板封口机和铝箔覆盖平板。

10)

在室温下孵育板30分钟。

11)

去除封口机和铝箔。立即加入100μL终止液到每个孔。在平板混合器上摇动板1分钟。

12)

用微量滴定板读数器在10分钟内读取450nm处的吸光度。

 

准备好所有的试剂和标准品

 

向每个孔中加入50μL测定稀释液

 

 

每孔加入50μLFGF-23标准品或样品

 

 

在平板混合器上在室温下孵育2小时

 

 

抽吸并洗涤4次

 

 

每孔加入100μL的FGF-23共轭物

 

 

在平板混合器上在室温下孵育1小时

Fibroblast growth factor-9, also called FGF-9, is an approximately 26 kDa secreted glycoprotein of the FGF family. Secreted human FGF-9 is a 205-207 aa protein that lacks the N-terminal 1-3 aa and shares 98% sequence identity with mouse, rat, equine, porcine and bovine FGF-9. In addition to FGF R3 (IIIb), FGF-9 binding to the IIIc splice forms of FGF R1, R2 and R3 are variably reported. FGF-9 is an autocrine/paracrine growth factor considered to be important for the growth and survival of motorneurons and prostate. Meanwhile, FGF9 is a potent mitogen and survival factor required for morphogenesis during embryonic development and numerous biological functions at adulthood. FGF9 have been shown to improve systolic function after myocardial infarction in a clinical trial. FGF9 promotes cardiac vascularization during embryonic development but is only weakly expressed in the adult heart.

Secreted human FGF-9 is a 205-207 aa protein that lacks the N-terminal 1-3 aa and shares 98% sequence identity with mouse, rat, equine, porcine and bovine FGF-9. In addition to FGF R3 (IIIb), FGF-9 binding to the IIIc splice forms of FGF R1, R2 and R3 are variably reported. FGF-9 is an autocrine/paracrine growth factor considered to be important for the growth and survival of motorneurons and prostate. Meanwhile, FGF9 is a potent mitogen and survival factor required for morphogenesis during embryonic development and numerous biological functions at adulthood. FGF9 have been shown to improve systolic function after myocardial infarction in a clinical trial. FGF9 promotes cardiac vascularization during embryonic development but is only weakly expressed in the adult heart.

Formulation

Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0 mg/mL. Stock solutions should be apportioned into working aliquots and stored at ≤ -20. Further dilutions should be made in appropriate buffered solutions.

This kit captures ADAMTS13-cleaved products using a sandwich method with the anti-GST mouse monoclonal antibody and the peroxidase-conjugated mouse monoclonal anti-N10 antibody. First, the anti-GST mouse monoclonal antibody immobilized on to the microplate reacts with the VWF73 substrate(GST-VWF73-His). The sample is then applied to the microplate, on which ADAMTS13 cleaves the VWF73 substrate. By applying the mouse monoclonal anti-N10 antibody conjugated with horseradish peroxidase(HRP conjugated antibody), the cleavage product is sandwiched between the two antibodies. Because the amount of the cleavage products depends on the ADAMTS13 activity, the amount of the enzyme-labeled antibodies that bind to the cleavage products also reflect the level of ADAMTS13 activity. Therefore, the plasma ADAMTS13 activity can be determined by colorimetrically measuring the amount of detached oxidized (colored)TMBZ using urea hydrogen peroxide(H2O2)and 3,3’,5,5’-Tetramethyl- benzidine(TMBZ)as the substrates.
