Fc gamma RIIIa is a low/intermediate affinity receptor for polyvalent immune-complexed IgG. It is involved in phagocytosis, secretion of enzymes and inflammatory mediators, antibody-dependent cytotoxicity and clearance of immune complexes. In humans, it is a 50-70 kDa type I transmembrane activating receptor expressed by NK cells, T cells, monocytes, and macrophages. Fc gamma RIIIb is highly related, sharing 97% amino acid (aa) identity within the extracellular domain (ECD), but is a GPI-linked receptor expressed on human neutrophils and eosinophils. The ECD of Fc gamma RIIIa shares 63%, 61%, 65%, 59% and 58% aa identity with mouse Fc gamma RIV, rat Fc gamma RIIIa, feline CD16, bovine CD16 and porcine Fc gamma RIIIb paralogs, respectively. The Fc gamma RIIIa cDNA encodes 254 aa including a 16 aa signal sequence, 191 aa ECD with two C2-type Ig-like domains and five potential N-glycosylation sites, a 22 aa transmembrane (TM) sequence and a 25 aa cytoplasmic domain.

正常人类细胞和动物细胞

Lifeline®人类细胞包括：内皮细胞，上皮细胞，成纤维细胞，造血细胞，黑素细胞，平滑肌和间充质干细胞。所有Lifeline®正常人和动物（小鼠）细胞：

当在生命线培养基中培养时，不需要暴露于抗微生物剂或酚红。不需要抗微生物剂和酚红，并且不推荐使用，以获得适的电池性能。这些补充剂可能导致细胞应激和“掩蔽效应”，可能会对实验结果产生负面影响。
支原体呈阴性，细菌和真菌生长呈阴性。
通过PCR对HIV，HBV和HCB呈阴性。

Receptors for the Fc region of IgG (Fc gamma Rs) are members of the Ig superfamily that function in the activation or inhibition of immune responses such as degranulation, phagocytosis, ADCC (antibody‑dependent cellular toxicity), cytokine release, and B cell proliferation. The Fc gamma Rs have been divided into three classes based on close relationships in their extracellular domains; these groups are designated Fc gamma RI (also known as CD64), Fc gamma RII (CD32), and Fc gamma RIII (CD16). Each group may be encoded by multiple genes and exist in different isoforms depending on species and cell type. The only inhibitory member in human and mouse, Fc gamma RIIb, has an intrinsic cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM). The coordinated functioning of activating and inhibitory receptors is necessary for successful initiation, amplification, and termination of immune responses.Mouse CD16 is encoded by a single gene. The protein product is a type I transmembrane protein having two extracellular Ig-like domains. It is expressed on a variety of myeloid and lymphoid cells and associates with Fc R gamma to deliver an activating signal upon ligand binding.

作用机制：

Clophosome^®-A（氯弗松），带负电荷的氯膦酸脂质体，是*代中性氯氟松Clophosome^®-N的升级产品，属于第二代的体内巨噬细胞清除剂（in vivo macrophage depletion），本质就是包裹氯膦酸分子的大单室脂质体（LUV）。结合游离氯膦酸不易穿透脂质体的磷脂双分子层和细胞膜，但脂质体可轻易被巨噬细胞吞噬的特点。氯氟松Clophosome被吞噬后，在溶酶体磷脂酶作用下，脂质体的磷脂双分子层被破坏，使得其内的水溶性药物–氯膦酸释放进入细胞。当氯膦酸在细胞内累积，且达到刚发挥药效的阈浓度（threshold concentration），细胞不可逆损伤，通过凋亡诱导进而死亡。而死后的巨噬细胞释放的游离氯膦酸，循环系统中半衰期极其短暂，泌尿系统循环中快速清除【见图1. 巨噬细胞自sha途径】。

如果脂质体能够充分注射进入体内，机体不同来源的巨噬细胞可被清除，包括肝脏（Kupffer cells，库普弗细胞），脾脏（不同的巨噬细胞亚群），肺脏（肺泡但不是小肠巨噬细胞），腹膜腔，淋巴结，关节（吞噬滑膜衬里细胞）和睾wan。
