肌动蛋白与细胞中的多种蛋白质相互作用
*，F-和G-肌动蛋白与细胞中的多种蛋白质相互作用（14）。目前有超过150种已知的肌动蛋白结合蛋白（ABP），其占细胞蛋白的约25％。ABP在肌动蛋白细胞骨架上赋予大量功能多样性，其涉及诸如肌肉收缩，唇形突破，细胞运动，胞质分裂和细胞质流动的过程。研究这些过程如何调节肌动蛋白丝功能和动力学是一个重要且不断增长的研究领域。

问题1：哪种肌动蛋白结合蛋白（ABP）可用作F-肌动蛋白结合对照？
答案1：肌动蛋白结合蛋白α-辅肌动蛋白（Cat。＃AT01）是一种F-肌动蛋白结合和交联蛋白，使其可用作F-肌动蛋白结合研究的对照。它的Kd是1μM，这意味着在该浓度下发生50％结合。因此，我们的目标是使用 2μM的α-肌动蛋白作为结合测定中的阳性对照。在结合反应中，2μM浓度的α-辅肌动蛋白相当于0.23μg/μl。此外，α-辅肌动蛋白对pH敏感，其高亲和力发生在pH 7.0。
问题2：Cytoskeleton的肌球蛋白或重的meromyosin是否具有F-肌动蛋白活力？
答案2：遗憾的是，我们不会出售运动级肌球蛋白或重肌红蛋白用于肌动蛋白运动试验。我们发现质量控制测试非常难以复制，因此我们无法充分确保试剂的质量。因此，肌球蛋白（Cat。＃MY02和MY03）和重型meromyosin蛋白（Cat。＃MH01）不推荐用于F-肌动蛋白运动测定。已经测试过这些试剂的客户在运动方面没有取得很好的成功，因此我们不建议购买样品进行测试。如果需要运动级肌动蛋白或重型meromyosin，我们建议遵循原始方案（Kron和Spudich，1985）并在液氮中冷冻100μl等分试样以确保稳定性。

纯度：
通过在SDS-PAGE凝胶上扫描蛋白质的光密度测定来确定纯度。样品纯度> 99％。
生物活性：

确定细胞群中丝状肌动蛋白（F-肌动蛋白）含量与游离球状肌动蛋白（G-肌动蛋白）含量的可重复和准确的方法是使用F-actin和G-actin细胞的Western印迹定量分数（1-4）。一般方法是使F-肌动蛋白稳定缓冲液中的细胞均质化，然后离心以将F-肌动蛋白与G-肌动蛋白池分离。然后通过SDS-PAGE分离级分，并通过Western印迹定量肌动蛋白。终结果给出了准确的方法来确定掺入细胞骨架的F-肌动蛋白与细胞质中发现的G-肌动蛋白的比例。该试剂盒包含进行该试验所需的所有试剂。

实施例结果
使用G-肌动蛋白/ F-肌动蛋白体内测定试剂盒，在用肌动蛋白聚合药物jasplakinolide处理的Swiss 3T3细胞中研究了G-肌动蛋白和F-肌动蛋白量的变化。在未经处理的瑞士3T3细胞中，80％的肌动蛋白是可溶性G-肌动蛋白，并且在上清液部分中发现，20％的肌动蛋白是丝状F-肌动蛋白并且在沉淀部分中发现。在用jasplakinolide处理的Swiss 3T3细胞中，80％的肌动蛋白重组成F-肌动蛋白并且在沉淀部分中发现（图1）。

问题1：在哪个步骤可以停止检测？
答案1： 直到在37℃下旋转100,000xg 1小时后才能停止测定。在高速离心后，可将上清液（G-肌动蛋白）与SDS上样缓冲液混合并冷冻备用。应使用解聚剂和水重悬沉淀（F-肌动蛋白），然后与SDS上样缓冲液混合并冷冻备用。在冷冻时，F-肌动蛋白解聚，因此在冷冻样品之前必须将F-肌动蛋白与G-肌动蛋白分离以分离样品以准确测量F-肌动蛋白和G-肌动蛋白比率。
问题2：这种检测有多敏感？
答案2： 该检测方法可以检测到G-肌动蛋白与F-肌动蛋白比例的15％变化。每个条件应一式两份进行并重复几次，因为实验之间的测定再现性可以变化10-20％。
