测定原理

APO LOGIX SR试剂盒含有一种通用的磺酰罗丹明标记的半胱天冬酶抑制剂（磺酰罗丹明 – 肽 – 氟甲基酮）。该试剂是可渗透细胞的，用于全细胞检测细胞凋亡。凋亡细胞通过荧光板阅读器或荧光显微镜在550nm处使用激发源检测，并在595nm处测量发射。该测定需要约1小时以完成

用星形孢菌素刺激Jurkat细胞2小时，然后用SR-VAD-FMK标记。

介绍

接触化学神经制剂，农药和某些药物（麻醉剂，KKY和治疗药物）会降低红细胞（RBC）乙酰胆碱酯酶（AChE）的活性。RBC-AChE可以作为一种生物标记物来监测外周和中枢神经系统中AChE功能的抑制和增加（9）。乙酰胆碱酯酶（AChE）是参与神经传递的zui重要的酶之一。该酶结合可兴奋组织（突触连接，内质网等）的细胞膜1-3。由于神经毒气和一类重要的农药对AChE的抑制，对人类和动物的急性毒性一直是一个深入的科学研究领域4,5。AChE抑制剂在临床上也被用作阿尔茨海默病的治疗药物（如他克林（四氢氨基吖啶））6，并且是各种疾病过程和治疗策略越来越受关注的对象7,8。然而，AChE抑制剂的环境检测和作为药物的AChE酶活性调节剂的开发已经由于当前测定方法的困难和复杂性而受到阻碍。

测定原理
