Beta actin also know as Cytoplasmic Actin is a 43 kDa, highly conserved protein, ubiquitously expressed in all eukaryotic cells. They comprise, along with microtubules, a major component of the cytoskeleton. β-Actin is a relatively stable cytoskeletal protein normally at a constant level in cells, regardless of experimental treatment or technical procedure. For this reason, measurement of β-Actin is generally used as an internal control for experimental error.

β-Actin Mouse Monoclonal Antibody -20℃保存,Western一抗稀释液-20℃或4℃保存,一年有效。Western一抗稀释液优先推荐4℃保存,长期不使用可以考虑-20℃保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。
注意事项:
如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验,请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体,包括已经使用过的稀释抗体,请4℃保存。 
回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10,000g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。
提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果,请考虑使用上述检测专用的一抗稀释液。

使用说明:
请根据抗体的实际用途选择相应的使用方法。
1. Western检测:

a. 按照推荐的稀释比例用碧云天提供的Western一抗稀释液稀释抗体。
b. 把经过封闭的蛋白膜与稀释好的一抗4℃缓慢摇动过夜或室温缓慢摇动2小时,确保稀释的抗体至少能在摇动的瞬间覆盖蛋
白膜。
c. 回收稀释的一抗,4℃保存以备下次继续使用。
d. 按照Western的实验步骤进行后续的洗涤、二抗孵育、洗涤和检测等操作。具体操作可以参考如下网页:

2. 免疫染色:

Background Beta actin also known as Cytoplasmic Actin is a 43 kDa, highly conserved protein, ubiquitously expressed in all eukaryotic cells. They comprise, along with microtubules, a major component of the cytoskeleton. β-Actin is a relatively stable cytoskeletal protein normally at a constant level in cells, regardless of experimental treatment or technical procedure. For this reason, measurement of β-Actin is generally used as an internal control for experimental error.

保存条件:

β-Actin Mouse Monoclonal Antibody (HRP Conjugated) -20℃保存,Western一抗稀释液-20℃或4℃保存,一年有效。Western一抗稀释液优先推荐4℃保存,长期不使用可以考虑-20℃保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。

注意事项:

如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验,请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体,包括已经使用过的稀释抗体,请4℃保存。

回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10,000g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。

提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果,请考虑使用上述检测专用的一抗稀释液。

1.Western检测:
a.按照推荐的稀释比例用碧云天提供的Western一抗稀释液稀释抗体。
b.把经过封闭的蛋白膜与稀释好的HRP一抗4℃缓慢摇动过夜或室温缓慢摇动2小时,确保稀释的抗体至少能在摇动的瞬间覆盖蛋白膜。
c.回收稀释的HRP一抗,4℃保存以备下次继续使用。
d.按照Western的实验步骤进行后续的洗涤和检测等操作,无须再孵育二抗。具体操作可以参考如下网页:

Background Beta actin also known as Cytoplasmic Actin is a 43 kDa, highly conserved protein, ubiquitously expressed in all eukaryotic cells. They comprise, along with microtubules, a major component of the cytoskeleton. β-Actin is a relatively stable cytoskeletal protein normally at a constant level in cells, regardless of experimental treatment or technical procedure. For this reason, measurement of β-Actin is generally used as an internal control for experimental error.

保存条件:

β-Actin Mouse Monoclonal Antibody -20℃保存,Western一抗稀释液-20℃或4℃保存,一年有效。Western一抗稀释液优先推荐4℃保存,长期不使用可以考虑-20℃保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。

注意事项:

如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验,请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体,包括已经使用过的稀释抗体,请4℃保存。

回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10,000g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。

提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果,请考虑使用上述检测专用的一抗稀释液。

1. Western检测:
a. 按照推荐的稀释比例用碧云天提供的Western一抗稀释液稀释抗体。
b. 把经过封闭的蛋白膜与稀释好的一抗4℃缓慢摇动过夜或室温缓慢摇动2小时,确保稀释的抗体至少能在摇动的瞬间覆盖蛋白膜。
c. 回收稀释的一抗,4℃保存以备下次继续使用。
d. 按照Western的实验步骤进行后续的洗涤、二抗孵育、洗涤和检测等操作。具体操作可以参考如下网页:

2. 免疫染色:

特异性磷抗体:Detection and quantitation of changes in the state of phosphorylation of specific proteins is of great utility in the quest to establish the function of a given protein and the consequences of its reversible phosphorylation. Two methods commonly used to measure protein phosphorylation and dephosphorylation in cell preparations employ prelabeling with 32Pi or back phosphorylation. These methods continue to be very effective and have advantages for many test systems, but they do have several practical and theoretical limitations (Nestler and Greengard, 1984). Based in large part on the successful use of short synthetic peptides to produce epitope-targeted antibodies (Lerner, 1982;Sutcliffe et al., 1983), an immunochemical approach became an attractive alternative for detecting changes in the state of phosphorylation of specific proteins at a specific site. The use of phosphorylation state-specific antibodies takes advantage of the sensitivity and selectivity afforded by immunochemical methodology, combined with relatively simple preparation and potentially broad applications.
The first report of phosphorylation-dependent antibodies appeared in 1981, when polyclonal antibodies that could detect phosphotyrosine-containing proteins were produced by immunization with benzyl phosphonate conjugated to keyhole limpet hemocyanin (KLH) (Ross et al., 1981). Shortly thereafter, Nairn and colleagues reported the production of serum antibodies that distinguished between the phospho- and dephospho-forms of G-substrate, a protein localized to cerebellar Purkinje cells and phosphorylated by cGMP-dependent protein kinase (Nairn et al., 1982). A synthetic heptapeptide, Arg-Lys-Asp-Thr-Pro-Ala-Leu, corresponding to a repeated sequence surrounding two phosphorylated threonyl residues in the intact protein, served as antigen. Rabbit antisera against a peptide-KLH conjugate were specific for the dephospho-form of G-substrate. Phospho-specific antibodies were prepared by immunization of rabbits with the purified phosphoprotein, phosphorylated in vitro to a stoichiometry of 2 mol/mol with cGMP-dependent protein kinase. Despite this initial success, other attempts in our laboratory to produce phospho-specific polyclonal antisera by immunization with the phospho-form of intact proteins were not very successful, probably because of two significant factors. First, many phosphorylated proteins are believed to undergo rapid dephosphorylation during immunization, regardless of the route of injection, leading to the loss of the desired phospho-epitope. Second, holoproteins generally contain multiple immunogenic epitopes; this decreases the probability that colonal dominance for a phospho-specific epitope will be obtained.
Taking a more direct approach utilizing phosphorylated and unphosphorylated forms of synthetic phosphopeptides, we developed a general protocol for the production of phosphorylation state-specific antibodies for substrates with established site(s) of phosphorylation (Czernik et al., 1991)). In early stages of our development of this methodology, phosphopeptides were routinely prepared by enzymatic phosphorylation (Czernik et al., 1991). Although this approach remains perfectly valid today, the preparation of synthetic phosphopeptides using Fmoc derivatives of phosphoamino acids has become the state-of-the-art (Czernik et al., 1995;Czernik et al., 1996). Likewise, we have examined the use of both polyclonal and monoclonal techniques for antibody production. Given the high success rate that we and others have obtained with the polyclonal technique, it has become the method of choice, because it is an easier and less costly method for the average laboratory. However, when appropriate, this approach can be readily adapted for monoclonal antibody production.

Q2。为什么SiR肌动蛋白(或微管蛋白)探针适用于STED显微镜检查?

A4。SiR探针在近红外/远红光谱范围内被激发并发光,因此避免使用较短波长,例如通常自发荧光的蓝光和绿光,从而产生更高的背景信号。SiR偶联探针具有两种物理状态:1。非荧光,封闭的关闭状态(螺内酯)和2.开放的,高度荧光的开启状态(两性离子)。探针与其配体靶的结合有利于高度荧光的开放状态,而游离的未结合的探针以闭合的非荧光状态存在(图4)。荧光扩增从未结合状态到结合状态是100倍。这导致高灵敏度的生物传感器,其中大部分荧光仅在结合状态下发生(参见参考文献3和4)。

Q5:SiR探针在室温下是否稳定?

A5:是的,探头在室温下稳定几天。但是,它很大程度上取决于探针和溶剂。因此,建议将所有探针或溶液储存在-20°C。

 

Q6:SiR-肌动蛋白和SiR-微管蛋白对细胞有毒吗?

本Actin抗体特异性识别α-smooth muscle actin,为进口分装,用人工合成的α-smooth muscle actin N-terminal一段十肽作为抗原制备而成的抗α-smooth muscle actin小鼠单克隆抗体。克隆号为1A4。本Actin抗体(smooth muscle specific)在Western blot或免疫染色时仅识别α-smooth muscle actin。
Actin即肌动蛋白,是细胞的一种重要骨架蛋白。同时Actin在细胞分泌、吞噬、locomotion、cytoplasmic  streaming和cytokinesis等过程中起重要作用。Actin在不同物种之间高度保守,以至于很难获得较好的针对actin的抗血清。Actin大致可分为六种,其中四种是不同肌肉组织特异性的,包括α-skeletal muscle actin,α-cardiac muscle actin,α-smooth muscle actin,和γ-smooth muscle actin;其余两种广泛分布于各种组织中,包括β-actin(β-non-muscle)和γ-non-muscle actin。不同的actin之间同源性大于90%,但在N-terminal同源性仅50-60%,因此N-terminal常被用作actin的抗原。
α-smooth muscle actin的免疫染色可以用于判断是否是smooth muscle。
配套提供了Western一抗稀释液,可以用于Western检测时的一抗稀释。
建议抗体使用时的稀释比例如下(实际使用时需根据抗原水平的高低作适当调整):

保存条件:
Actin抗体(smooth muscle specific)-20℃保存,Western一抗稀释液-20℃或4℃保存,一年有效。Western一抗稀释液优先推荐4℃保存,长期不使用可以考虑-20℃保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。
注意事项: 
在Western实验后,请注意回收稀释的抗体。回收的抗体在进行Western实验时至少可以重复使用10次。稀释后的抗体,包括已经使用过的稀释抗体,4℃保存。
回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10000g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。
本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品,不得存放于普通住宅内。

b. 把经过封闭的蛋白膜与稀释好的一抗4℃缓慢摇动过夜或室温缓慢摇动1-2小时,确保稀释的抗体至少能在摇动的瞬间覆盖蛋白膜。
c. 回收稀释的一抗,4℃保存以备下次继续使用。
d. 按照Western的实验步骤进行后续的洗涤、二抗孵育、洗涤和检测等操作。具体操作可以参考如下网页: http://www.beyotime.com/support/western.htm
2. 免疫染色:

Background Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. This antibody is predicted to react with all actin isoforms, including: 1) sarcomeric actins, α-skeletal (α-SKA) and α-cardiac (α-CAA), 2) smooth muscle actins (SMAs), α-SMA and γ-SMA, and 3) cytoplasmic actins (CYAs), β-CYA and γ-CYA.

保存条件:

Pan-Actin Rabbit Monoclonal Antibody -20℃保存,需避免反复冻融。Western一抗稀释液-20℃或4℃保存,一年有效,Western一抗稀释液优先推荐4℃保存,长期不使用可以考虑-20℃保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。

注意事项:

如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验,请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体,包括已经使用过的稀释抗体,请4℃保存。

回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10,000×g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。

提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果,请考虑使用上述检测专用的一抗稀释液。

使用说明:
请根据抗体的实际用途选择相应的使用方法。
1.Western检测:
a.按照推荐的稀释比例用碧云天提供的Western一抗稀释液稀释抗体。
b.把经过封闭的蛋白膜与稀释好的一抗4℃缓慢摇动过夜或室温缓慢摇动2小时,确保稀释的抗体至少能在摇动的瞬间覆盖蛋白膜。
c.回收稀释的一抗,4℃保存以备下次继续使用。
d.按照Western的实验步骤进行后续的洗涤、二抗孵育、洗涤和检测等操作。具体操作可以参考如下网页:

2. 免疫染色:
