本辣根过氧化物酶标记山羊抗大鼠IgG(H+L)(HRP-labeled Goat Anti-Rat IgG(H+L))为进口分装,用于Western、ELISA和IHC等的二抗。HRP,即horseradish peroxidase,中文名为辣根过氧化物酶,可以在Western检测时催化ECL类试剂(如ECL、BeyoECL等)产生化学发光,可以在ELISA时催化TMB产生蓝色,也可以在IHC或Western blot检测时催化DAB产生棕色沉淀。
本辣根过氧化物酶标记山羊抗大鼠IgG(H+L)用于WB、ELISA和IHC的推荐稀释比例参考上表,实际实验操作过程中需根据抗原和抗体的具体情况适当调节二抗的稀释比例。例如对于WB,推荐的可以调节的稀释范围为1:500-5000。
本抗体为用纯化的大鼠IgG免疫山羊,然后用亲和纯化柱对获得的抗血清进行纯化而获得的优质二抗。
本抗体如果用于常规的Western检测,以每次检测需10毫升1:1000稀释的二抗计,至少可以检测50次。

本辣根过氧化物酶标记Streptavidin (HRP-labeled Streptavidin),也称HRP-Streptavidin、Streptavidin-HRP或辣根过氧化物酶标记链霉亲和素,可以用于生物素(Biotin)标记的抗体、核酸、蛋白或其它生物素标记分子的检测。具体用途包括Western、ELISA、免疫组化或免疫细胞化学、原位杂交、Southern、Northern、EMSA等。

Streptavidin的分子量为52kD,可以高度特异性地和生物素(Biotin)结合。Streptavidin和生物素的亲和常数为Kd=10-15M。Streptavidin是一个4聚体蛋白,可以同时结合4个生物素分子。Streptavidin的中文名为链霉亲和素,从Streptomyces adidinii中纯化获得,和鸡蛋清来源的Avidin(亲和素)在空间结构以及与生物素的亲和力方面具有高度的相似性。和Avidin不同的是,Streptavidin是一种非糖基化蛋白,并且基本不带电荷。Avidin的pI= ~10.5,在中性pH条件下呈碱性。由于Streptavidin和Avidin相比在中性条件下不带电荷,因此Streptavidin的非特异性结合比Avidin低很多,这样检测时的非特异性背景就低很多。因此目前在生物素检测时通常使用Streptavidin替代Avidin [1]。

辣根过氧化物酶即horseradish peroxidase (HRP),可以在Western、EMSA、Southern或Northern等检测时催化ECL类试剂(如ECL、BeyoECL等)产生化学发光,可以在ELISA时催化TMB产生蓝色,也可以在免疫组化、免疫细胞化学或Western blot检测时催化DAB产生棕色沉淀[2]。

本HRP-Streptavidin用高纯度的Streptavidin和超纯的辣根过氧化物酶交联而成,确保产生最低的本底和最高的灵敏度。不同浓度生物素标记二抗使用本产品进行显色检测效果参见图1。

图1. 碧云天辣根过氧化物酶标记Streptavidin (A0305)的检测效果图。使用兔抗山羊抗体对96孔板进行包被和封闭后,将生物素标记山羊抗小鼠IgG(H+L) (A0286)稀释至图中所示各浓度,加入各反应孔中室温孵育1小时;洗板后,分别加入100ng/ml的辣根过氧化物酶标记Streptavidin本产品(A0305)和国外T公司(Competitor)的同类产品,室温孵育25分钟;洗板后,加入100µl TMB显色液(ELISA HRP显色用) (P0209),室温避光孵育5分钟后加入TMB显色终止液(450nm, 不含硫酸) (P0215)并在450nm检测吸光度。图A为使用本产品和国外T公司同类产品进行检测的标准曲线,生物素标记二抗浓度为横坐标,A450为纵坐标。图B为检测低浓度的生物素标记二抗的吸光度。图中可见,在比较适宜的生物素标记样品浓度范围内,本产品的检测信号比国外T公司同类产品高约1倍。实测数据会因试剂、检测仪器等的不同而存在差异,图中数据仅供参考。

本辣根过氧化物酶标记Streptavidin的浓度为1mg/ml。本产品用于各种常规用途的推荐稀释比例参考下表,实际实验操作过程中需根据具体情况适当调节辣根过氧化物酶标记Streptavidin的稀释比例。对于Western,推荐的稀释范围为1:2000-10,000。

本辣根过氧化物酶标记Streptavidin (HRP-labeled Streptavidin),也称HRP-Streptavidin、Streptavidin-HRP或辣根过氧化物酶标记链霉亲和素,可以用于生物素(Biotin)标记的抗体、核酸、蛋白或其它生物素标记分子的检测。具体用途包括Western、ELISA、免疫组化或免疫细胞化学、原位杂交、Southern、Northern、EMSA等。

Streptavidin的分子量为52kD,可以高度特异性地和生物素(Biotin)结合。Streptavidin和生物素的亲和常数为Kd=10-15M。Streptavidin是一个4聚体蛋白,可以同时结合4个生物素分子。Streptavidin的中文名为链霉亲和素,从Streptomyces adidinii中纯化获得,和鸡蛋清来源的Avidin(亲和素)在空间结构以及与生物素的亲和力方面具有高度的相似性。和Avidin不同的是,Streptavidin是一种非糖基化蛋白,并且基本不带电荷。Avidin的pI= ~10.5,在中性pH条件下呈碱性。由于Streptavidin和Avidin相比在中性条件下不带电荷,因此Streptavidin的非特异性结合比Avidin低很多,这样检测时的非特异性背景就低很多。因此目前在生物素检测时通常使用Streptavidin替代Avidin [1]。

辣根过氧化物酶即horseradish peroxidase (HRP),可以在Western、EMSA、Southern或Northern等检测时催化ECL类试剂(如ECL、BeyoECL等)产生化学发光,可以在ELISA时催化TMB产生蓝色,也可以在免疫组化、免疫细胞化学或Western blot检测时催化DAB产生棕色沉淀[2]。

本HRP-Streptavidin用高纯度的Streptavidin和超纯的辣根过氧化物酶交联而成,确保产生最低的本底和最高的灵敏度。不同浓度生物素标记二抗使用本产品进行显色检测效果参见图1。

本辣根过氧化物酶标记Streptavidin的浓度为1mg/ml。本产品用于各种常规用途的推荐稀释比例参考下表,实际实验操作过程中需根据具体情况适当调节辣根过氧化物酶标记Streptavidin的稀释比例。对于Western,推荐的稀释范围为1:2000-10,000。

本辣根过氧化物酶标记山羊抗人IgG (H+L) (HRP-labeled Goat Anti-Human IgG (H+L))为进口分装,用于Western、ELISA和IHC等的二抗。HRP,即horseradish peroxidase,中文名为辣根过氧化物酶,可以在Western检测时催化ECL类试剂(如 ECL、BeyoECL等)产生化学发光,可以在ELISA时催化TMB产生蓝色,也可以在IHC或Western blot检测时催化DAB产生 棕色沉淀。
本辣根过氧化物酶标记山羊抗人IgG (H+L)用于WB、ELISA和IHC的推荐稀释比例参考上表,实际实验操作过程中需根据 抗原和抗体的具体情况适当调节二抗的稀释比例。对于WB,推荐的稀释范围为1:500-2000。
本抗体为用纯化的人IgG免疫山羊,然后用亲和纯化柱对获得的抗血清进行纯化而获得的优质二抗。
本抗体如果用于常规的Western检测,以每次检测需10毫升1:1000稀释的二抗计,至少可以检测100次。

本Biotin标记辣根过氧化物酶 (Biotin-labeled horseradish peroxidase),也称生物素标记辣根过氧化物酶、生物素标记过氧化物酶、Biotin-labeled HRP、Biotin-HRP、biotinylated peroxidase或辣根过氧化物酶标记生物素,为进口分装,可以用于链霉亲和素(Streptavidin)和亲和素(avidin)或其标记或结合的抗体、核酸、蛋白等的检测。具体用途包括Western、ELISA、免疫组化或免疫细胞化学、原位杂交、Southern、Northern、EMSA等。

本Biotin-HRP用高纯度的Biotin和超纯的辣根过氧化物酶(RZ>3)通过化学键偶联而成,确保产生极低的本底和极高的灵敏度。本产品中生物素标记辣根过氧化物酶的蛋白浓度为1mg/ml。

生物素(Biotin)又称维生素H、辅酶R,是水溶性维生素,也属于B族维生素,CAS号为58-85-5,分子式为C10H16N2O3S,分子量为244.31。Avidin或Streptavidin是一种4聚体蛋白,可以同时高度特异性地结合4个生物素分子,是一种天然存在生物分子相互作用体系。生物素与Avidin或Streptavidin的结合系统,被称为生物素-亲和素或生物素-链霉亲和素系统,该系统是一种具有高度亲和力、高灵敏度、特异性强和稳定性好等优点的信号放大标记技术,已广泛应用于多种检测系统,如免疫检测、DNA杂交检测、免疫组化、流式细胞术、目标分子的纯化和表征等,有利于信号放大和增加灵敏度。

辣根过氧化物酶(horseradish peroxidase, HRP),可以在Western、EMSA、Southern或Northern等检测时催化发光底物试剂(如ECL、BeyoECL等)产生化学发光,可以在ELISA检测时催化TMB产生蓝色,也可以在免疫组化、免疫细胞化学或Western blot检测时催化DAB产生棕色沉淀。

本Biotin标记辣根过氧化物酶用于各种常规用途的推荐稀释比例参考下表,实际实验操作过程中需根据具体情况适当调节Biotin标记辣根过氧化物酶的稀释比例。对于Western,推荐的稀释范围为1:2000-10,000。
