BirA 生物素-蛋白质连接酶 (EC 6.3.4.15) 通过 ATP 中间体(生物素 5'-腺苷酸)以高效且有针对性的方式将 d-生物素与生物素受体肽/蛋白质共价添加。酶促生物素化蛋白质的下游应用是多种多样的、重要的和强大的。生物素-亲和素/链霉亲和素相互作用经常被用于亲和层析或蛋白质固定在表面或底物上。通过抗生物素抗体或亲和素/链霉亲和素-报告酶偶联物(-HRP、-碱性磷酸酶)或荧光探针进行蛋白质检测成为可能。生物素化 MHC 分子的多聚体形式是免疫生物学中的常用工具。

当与我们的 AviTag™ 生物素受体肽氨基酸序列结合使用时,生物素化发生的速度是天然大肠杆菌 BCCP 底物的两倍,与其他肽序列的生物素化相比,其发生率高出一个数量级或更多。因此,与其他可用序列相比,AviTag™ 序列需要更少的 BirA 酶和更短的孵育时间来完成生物素化。如果蛋白质不稳定性或蛋白酶活性是一个问题,这可能对下游的成功很重要。

Centrifuge the tube before opening. Reconstituting to a concentration more than 100 μg/ml is recommended. Dissolve the lyophilized protein in distilled water.

2-7 days, 2 ~ 8 °C under sterile conditions after reconstitution.

3 -6 months, -85~-65℃ under sterile conditions after reconstitution.

Recommend to aliquot the protein into smaller quantities when first used and avoid repeated freeze-thaw cycles.

重构方法(Reconstitution)

Centrifuge the tube before opening. Reconstituting to a concentration more than 100 μg/ml is recommended. Dissolve the lyophilized protein in distilled water.

The product should be stored at -25~-15℃ for 1 year from date of receipt.

2-7 days, 2 ~ 8 °C under sterile conditions after reconstitution.

3 -6 months, -85~-65℃ under sterile conditions after reconstitution.

Recommend to aliquot the protein into smaller quantities when first used and avoid repeated freeze-thaw cycles.
