 基本介绍： 霍乱毒素（Cholera toxin，CTX）是霍乱弧菌产生的毒力因子，会引起严重腹泻以及人体脱水。CTX是一种分子量约84kDa的 寡蛋白，包含一个A亚基以及包围在周围的5个B亚基构成。CTX是一种有效的腺苷酸环化酶（Adenylate cyclase，AC）活化 剂，也是负责霍乱症状的病原物质。霍乱毒素B亚基（a.k.a. 霍乱类毒素choleragenoid）负责全毒素识别并结合到哺乳动物细 胞表面的GM1神经节苷脂受体，促进A亚基进入细胞；A亚基承担ADP核糖转移酶（ADP-ribosyl-transferase）活性，此酶能 下调Gs蛋白表达并活化AC酶，进而促进cAMP水平提高。A亚基还能ADP核糖基化视杆细胞外节膜盘的转运蛋白，使得GTPase 失活。归因于GM1神经节苷脂受体普遍存在于真核细胞膜上，CTX用于各种各样的模型系统中活化腺苷酸环化酶（AC）。CTX 还是一种的黏膜疫苗佐剂，通过抑制IL-12生产来诱导2型辅助性T细胞的免疫应答。

基本介绍： 百日咳毒素（Pertussis toxin，PTX）是一种由6个非共价连接亚基组成的多元件蛋白，分子量范围约为9-28kDa，这些亚基分别命名为 S1，S2，S3，S4和S5，天然百日咳毒素中所占比为1:1:1:2:1，其中S4亚基以2倍形式存在。大亚基为S1（也称为A启动子），包含 NAD+糖基水解酶和ADP-核糖基转移酶活性。单独的A启动子能够将NAD+的ADP核糖转移到Gαi，Gαo或Gαt家族G蛋白的α亚基。PTX的结 晶结构显示其呈金字塔形状，A启动子位于塔尖，中间是S5，底端是二个二聚体，分别是S2-S4,和S3-S4。这5个亚基统称为B寡聚体亚基 （S2，S3，S4，S4和S5），允许PTX进入大部分细胞，使其粘附在包括TLR4和糖蛋白Ib在内受体蛋白的多糖残基上。通过受体介导的内 吞作用进入细胞后，PTX通过胞内体途径和高尔基体复合物逆向转运进入内质网。A启动子从PTX上释放出来，穿透内质网膜并导致目标 膜偶联的G蛋白失活。

炭疽毒素Anthrax Toxins

 

炭疽杆菌是炭疽病的病原体。炭疽杆菌产生的主要毒力因子是γ连接的聚-D-谷氨酸胶囊和由三种成分组成的外毒素，保护性抗原（PA），致死因子（LF）和水肿因子（EF）。这些蛋白质不是单*毒的，而是以二元组合起作用。PA（细胞结合成分）与LF（酶部分之一）的复合物被称为“致死毒素”并可导致死亡。PA和酶促EF一起引起皮肤水肿。分泌后，PA被膜肽酶切割，使63kDa羧基末端片段寡聚化成七聚体。切割是暴露EF和LF的结合位点的必要步骤。组装的PA复合物和相关的LF或EF通过胞吞作用进入细胞。PA介导LF和EF向细胞质的转移，其中这些酶识别并蛋白水解其靶标。LF组分是一种锌依赖性金属蛋白酶，可裂解丝裂原活化蛋白激酶激酶（MAPKK），从而削弱其功能，从而阻断MAPK信号通路。EF是钙调蛋白依赖性腺苷酸环化酶，其在细胞中产生过量的环AMP，导致水肿。目前的研究集中于开发有效的疫苗以及筛选LF酶活性的有效抑制剂。
