转染试剂线性PEI溶液的配制（1mg/ml）

聚乙烯亚胺（Polyethylenimine，PEI）是聚乙烯亚胺是一种具有较高的阳离子电荷密度的有机大分子，每相隔二个碳个原子，即每“第三个原子都是质子化的氨基氮原子，使得聚合物网络在任何pH下都能充当有效的“质子海绵”(proton sponge)体。PEI，可用作转染试剂，它可以缩聚DNA分子形成颗粒并转染真核细胞。有实验证明，分子量为25000的线性PEI即Polysciences 23966转染效率zui高。

下面将主要介绍polysciences 线性PEI(#23966)的产品特性及其溶液的配制过程。

2. 溶液配制

此指导说明书是经polysciences公司研究，测试及其客户反馈的后编写的。下面推荐一种简单适用而的方法，讲述如何配制浓度为1mg/mL的转染试剂的配制。

材料：

1g #23966、1L 纯水（Milli-Q®纯水，注射用水（WFI），或类似的生物级水）、12 M盐酸、10 M氢氧化钠、一次性0.1-0.2µm PES真空无菌过滤器、无菌高聚乙烯或聚丙烯试剂瓶。

器材：

1 L 玻璃烧杯、1 L玻璃量筒、核准pH计、2支1 mL塑料移液管、托盘、聚四氟乙烯（PTFE）搅拌棒、真空泵

配制过程：

1g #23966——900mL水溶解——搅拌、加热（60-80°C）——加HCl调pH到2.0左右——盖上烧杯，搅拌3h（直至粉末*溶解）——冷却至室温——加NaOH调pH到6.9-7.1——移液到量筒，加水补足至1L——真空过滤消毒——分装，-20°C保存。

备注：加热或加酸都有助于PEI溶解。当然酸可能不用加那么多，详细用量可试加一点搅拌看看，足够溶解就行，不一定需要加到pH那么低。

试剂储存：

冷冻部分溶液可在-20°C保存长达一年；部分解冻的溶液可在4°C保存2周，不可反复冻融。

粉末储存：

未开封的粉末有效期两年。可在室温下保存直至使用。

转染操作流程：(瞬时转染方法) 转染效率（部分数据参考，本数据来源于网络，不作为售后投诉依据）

1. 接种细胞： 转染前一天，用胶原酶（WORTHINGTONG 公司 LS004196)消化细

胞并计数（不建议用胰酶）。调整细胞浓度，将细胞铺入细胞培养的器皿，每个孔置

入的细胞量应能使转染时细胞汇合度达到 70~80%。

2. 准备 DNA-PEI 复合物： DNA、PEI 试剂和稀释剂在进行以下步骤前需先使其

升至室温。，用 Opti-MEM ITM（Invitrogen）或其他适合的无蛋白培养基稀释适

量 DNA。用同样的培养基稀释 PEI 试剂。每 1 μg DNA 需用 1-5 μL 线性 PEI 转

染试剂。一边轻轻涡旋装有 DNA 溶液 的试管，一边将稀释的线性 PEI 转染试剂滴

加至试管中（注意：请勿颠倒添加顺序）。充分混匀后，室温静置 10~25 min 以

形成 DNA-PEI复合物。当溶液体积较大时，请用圆底聚丙烯管，例如 NEST 5 mL /14

mL 离心管。

3. 转染细胞： 直接向每个孔中加入 DNA-PEI 复合物并轻轻涡旋培养板/培养皿。在

无血清条件下转染时，去除生长培养基，替换成无血清培养基，然后滴 DNA-PEI 复

合物。转染 3 h 后，添加½体积的包含 30%血清的生长培养基。

4. 孵育细胞和分析结果： 在 CO2 培养箱中 37℃下孵育细胞至可以分析检测。转

染后快 的话7 h 即可检测到转入基因的表达。 请自行确定适合检测时间。

Background:

Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one of which utilizes NAD(+) as the electron acceptor and the other NADP(+). The IDH2 homodimer is an the NADP(+)-dependent isocitrate dehydrogenase found in the mitochondria. It plays a role in intermediary metabolism and energy production and may tightly associate or interact with the pyruvate dehydrogenase complex.

Human isocitrate dehydrogenase 2, also known as IDH2, GenBank Accession No. NM_002168, a.a. 1-452(end) with with Arg to Gln mutation on a.a. 140 and C-terminal FLAG-tag, MW = 52 kDa, expressed in a baculovirus infected Sf9 cell expression system.

IDH reductive activity was measured in 200 µl reaction containing 25 mM Tris (pH 7.4), 150 mM NaCl, 10 mM MgCl2, 0.03% BSA, 1 mM alpha-Ketoglutarate, 10 µM NADPH and IDH. Depletion of NADPH was monitored continuously at Abs340 nm for 20 min. Molar extinction coefficient of NADPH is 6,200 M-1cm-1.

Lucerna——活体胞内RNA示踪

绿色荧光蛋白的发现及其应用是生命科学研究*一个重要的里程碑，使得细胞内不可见的重要蛋白无所遁形，进而明确其功能与调控机制。相较于蛋白质，近年在细胞内还揭示了一大类重要的分子——非编码RNA，这类特殊的RNA不编码蛋白，但是其数目远较编码RNA（mRNAs）多，包括microRNAs, Piwi-interacting RNAs, termini-associated RNAs等。目前对这一类RNA的研究方兴未艾，因技术上缺乏关键的研究工具对其细胞内定位于功能了解知之甚少。

胞内RNA成像技术可能是非编码RNA研究领域取得突破的关键，与绿色荧光蛋白相似，在目的RNA上标记荧光分子可以使RNA在荧光显微镜下呈现，明确其胞内定位，进而通过分子互作或药理学等技术了解其功能。然而目前的常规RNA成像技术难以在活细胞上应用，并且有效性存在很大问题。针对这一难题，来自威斯康星大学的研究人员成立了Lucerna公司，并研发了一种*的基因编码系统用于活细胞内RNA的荧光标记。该系统利用一种可与绿色荧光蛋白媲美的RNA原件——Spinach（一种RNA与源于绿色荧光蛋白的荧光小分子结合体，可与目的RNA结合并发射荧光）使得在活细胞中实时成像目的RNA的动态机制。利用该系统，研究人员可望在RNA胞内转运，降解、毒性RNA聚焦以及其他RNA调控机制的研究中取得突破性进展。
