Q: UTP的消光系数为10,000 Lmol-1cm-1 at 262 nm,而N1-甲基假UTP的消光系数为8,877 Lmol-1cm-1 at 271 nm,二者差别较大,在用N1-甲基假UTP替代UTP转录的mRNA A260/A280小于2.0,原有的mRNA浓度计算公式RNA浓度(ng/uL) =0D260xN(稀释倍数)x40就不在适用了,请问浓度计算公式系数40变为多少合适?有相关研究数据或文献资料分享吗?
A: 目前没有了解过是否影响计算,NanoDrop 的检测方法比较粗略,如果想要更精确的检测,可以使用Qubit检测RNA的浓度,这两种方法也是市面上比较常见的两种检测方法。
Q: 由于之前使用的其他公司的UTP,想要用翌圣的原料去替换,是否使用方法不同?
A: 一般采用完全替代的方式,假尿苷或者 N1-甲基假尿苷的使用量与原体系尿苷的使用量一致。
