固定剂、破膜剂用于细胞内细胞因子染色前对细胞膜的处理。固定剂起稳定细胞膜、保持细胞膜表面抗体与抗原结合的作用;破膜剂使流动的、完整的细胞膜产生小孔以利于抗体进入细胞。用流式细胞仪检测细胞内抗原及DNA时,对待测细胞进行固定、破膜处理后,加入相应抗体或检测试剂,孵育一定时间即可上机分析。   

产品组分

4%多聚甲醛、NaOH、NaH2PO4 、1%胎牛血清;pH 7.4-7.6

运输与保存方法

冰袋(wet ice)运输。

4℃避光保存,勿冰冻。有效期一年

注意事项

1)固定剂中含有有害物质甲醛溶液,使用时避免与皮肤、眼睛及黏膜接触。
2)本产品仅作科研用途!

应用实例

1. 收获待测细胞,1000 rpm离心10 分钟,弃上清。

2. 加入 500 uL预冷(4℃)固定剂。

3. 室温孵育10 分钟。

4. 1000 rpm离心10 分钟。

5. 弃上清,加入PBS洗一次,1000 rpm离心10 分钟。

6. 弃上清,加入1.5 mL破膜剂。

7. 室温孵育10-15 分钟。

8. 1000 rpm离心5 分钟。

9. 弃上清,加入适量破膜剂重悬细胞。

10. 加入检测抗体进行染色后,用流式细胞仪进行分析。
