5×SDS-PAGE蛋白上样缓冲液(SDS-PAGE Sample Loading Buffer),是一种经过改良的以溴酚蓝为染料,5倍浓缩的蛋白上样缓冲液。

SDS可与蛋白质结合使蛋白质-SDS复合物上带有大量的负电荷, 这时蛋白质本身的电荷完全SDS掩盖,消除了各种蛋白质本身电荷的差异, SDS还可以断开分子内和分子间的氢键,破坏蛋白质分子的二级和三级结构, DTT可以断开半胱氨酸残基之间的二硫键, 破坏蛋白质的空间结构, 消除了蛋白结构之间的差异。因此,电泳速度只是与其分子量大小有关。溴酚蓝用作电泳时的指示剂, 可大概指示电泳结束的时间。

可以用于常规的SDS-PAGE蛋白样品的上样。

注意事项

1)为了您的安全和健康,请穿实验服并戴一次性手套操作。

2)5×SDS-PAGE蛋白上样缓冲液中含少量DTT,有轻微刺激性气味,但不含剧毒的巯基乙醇。

3)5×SDS-PAGE蛋白上样缓冲液必须完全溶解后再使用。

 

操作方法

1)在室温或不超过37的水浴中溶解5×SDS-PAGE上样缓冲液。水浴溶解后立即室温存放, 尽量避免长时间置于水浴中。

2)按照每4 μL蛋白样品加入1 μL 5×SDS-PAGE蛋白上样缓冲液的比例, 混合蛋白样品和5×SDS-PAGE蛋白上样缓冲液。

3)100℃或沸水浴加热3-5分钟,以充分变性蛋白。

4)冷却到室温后,直接上样到SDS-PAGE胶加样孔内即可。

5)通常电泳至蓝色染料到达胶的底端处附近即可停止电泳。
